Abstract

Process Analytical Technology (PAT) is increasingly shaping pharmaceutical manufacturing by moving quality assurance from traditional end-product testing toward real-time process understanding. Through the integration of advanced analytical tools within manufacturing operations, PAT enables continuous monitoring and control of critical material attributes and process parameters during production. This review examines the fundamental concepts of PAT and its close alignment with Quality by Design (QbD) and risk-based manufacturing approaches. Key PAT techniques-including spectroscopic, imaging, acoustic, and mass-based methods are discussed in relation to major pharmaceutical unit operations such as blending, granulation, drying, coating, and tableting. In addition, the regulatory framework supporting PAT adoption is reviewed, with particular emphasis on ICH Q8(R2), Q9(R1), Q10, Q11, Q12, and Q13 guidelines, which encourage enhanced process understanding, effective lifecycle management, and the implementation of continuous manufacturing. While PAT offers significant advantages, including improved process robustness, reduced variability, real-time release capability, and increased manufacturing efficiency, challenges related to data management, sensor integration, and model reliability remain. Overall, PAT represents a key step toward more transparent, predictable, and scientifically driven pharmaceutical manufacturing.
