Abstract

Corn (Zea mays L.) is a food crop in Indonesia, one of the factors of decreased production is plant diseases, one of which is sheath blight caused by Rhizoctonia solani. The purpose of this study was to identify corn sheath blight caused by Rhizoctonia solani and to assess the pathogenicity and virulence of Rhizoctonia solani isolates. The research methods used included the exploration stage of sheath blight disease in several areas in Central Java, macroscopic and microscopic morphological identification, molecular identification using specific primers, and pathogenicity tests. The results of the study showed that, at the exploration stage, the symptoms observed included widespread tissue damage in the form of lesions and the presence of sclerotia. Morphological identification of the mycelial distribution pattern was scarce, the sclerotia distribution pattern was on the peripheral and scattered, the branching angle was 90º, and in the multinucleate category. Molecular identification showed that sheath blight was caused by Rhizoctonia solani AG1-IA. Pathogenicity test showed the fastest incubation period in isolates from Boyolali, at 5,80 days after inoculation; incidence reached 100% at week 4; the highest intensity in isolates from Boyolali was 73%. The conclusion is the pathogen causing corn sheath blight in Central Java is Rhizoctonia solani AG1-IA, and the pathogenicity test results showed that the Boyolali isolate had the highest intensity and incubation period.
